Figure 3 PD1 and TIM3 blockade restores IL10 and IFN gamma antibacterial immunity in AAH. (A) PBMC from 8 AAH were preincubated ± anti-PD1 and anti-TIM3 blocking antibodies, stimulated with LPS and assessed for IFN gamma+ and IL10+ PBMC by ELISpot. Blockade of PD1 and TIM3 increased IFN gamma+ PBMC, reduced IL10+ PBMC and improved the IFN gamma/IL10 ratio in AAH but not in alcohol-related cirrhosis (ARC), healthy controls (HC), and chronic hepatitis B cirrhotics. (B) Protein levels in 4 representative ELISpot supernatants from AAH were also modulated by the blocking strategy with increased IFN gamma and decreased IL10. (C) Total numbers of LPS-challenged IL10-producing cells ± anti-PD1 and anti-TIM3 blocking antibodies were lower in all immune subsets apart from monocytes. (D) Blockade of PD1 and TIM3 significantly augmented neutrophil antibacterial functions (oxidative burst to E coli, left; ingestion capacity, right) in 6 AAH.
