Fig. 1 UPGE2V and U6-keto-PGF1αV in cirrhotic rats with ascites under baseline conditions (□) and 60 minutes after administration of COX inhibitors (▨; ketorolac, n = 8; SC-236, n = 9). Values represent means ± SEM. Data are compared by the Studentt test for paired data. Fig. 2 Percent changes in V, UNaV, GFR, RPF, and UPGE2V induced by SC-236 (□; n = 12) or ketorolac (▨; n = 10) in cirrhotic rats with ascites. Values represent means ± SEM and correspond to the changes observed 60 minutes after the administration of COX inhibitors. Data are compared by the Student t test for unpaired data. Fig. 3 Ex vivo TXB2 production under baseline conditions (□) and after administration of COX inhibitors (▨) to rats with cirrhosis and ascites (ketorolac, n = 5; SC-236, n = 5). Whole blood samples were exposed to ionophore A23187 (10 minutes, 37°C), and the incubations were terminated by addition of cold methanol. TXB2 levels were determined in unextracted samples by ELISA. Results are given as means ± SEM. Data are compared by the Student t test for paired data. Fig. 4 COX-1 and COX-2 mRNA expression in renal tissue isolated from control and cirrhotic rats with ascites. Total RNA was subjected to RT-PCR using specific oligonucleotides for COX-1, COX-2, and GAPDH. RNA isolated from Raw macrophages was used as a positive control (first lane at right). These agarose gels are representative of results obtained from 3 different experiments. Fig. 5 Effects of ketorolac (n = 15) and SC-236 (n = 13) on renal water metabolism in rats with cirrhosis and ascites in protocol 2. (A) Percent water load excreted and (B) mUOsm were measured under baseline conditions (□) and after administration of COX inhibitors (▨) to cirrhotic rats. Results represent means ± SEM. Data are compared by the Student t test for paired data. Gastroenterology , DOI: ( /S (99)70020-X) Copyright © 1999 American Gastroenterological Association Terms and Conditions
