Figure 1 Signal transduction paradigm for cell proliferation. Cellular mitogenic stimulus can induce the autophosphorylation and activation of intrinsic tyrosine kinase activity of specific membrane receptors, which, in turn, can sequentially activate Ras, Raf, and MAP kinase. The activated cytoplasmic MAP kinase translocates into the nucleus and activates various transcription factors and proto-oncogenes associated with cell proliferation. PKC and G-protein–mediated events can also activate MAP kinase signaling. Atherogenic lipoproteins, by activating specific membrane receptors, may induce the activation of Ras-MAP kinase signaling.
