2 Figure 1 (A) Total RNA from oviducts was isolated with Trizol reagent (Invitrogen), and complementary DNA was then synthesized from 2 μg total RNA in 20 μL reaction mixture according to standard procedures. Complementary DNA obtained from oviduct with indicated genotype was amplified with primers for Lgr4 and arbp. Arbp was used as an endogenous control. Primer sets were described previously (9). (B) Mature (10-14 weeks) and natural-ovulated (P<.0005) or immature (4 weeks) and superovulated (P<.05) mice were mated with C57BL/6J male mice, and the average numbers of offspring are shown. To induce superovulation, female mice (4 weeks) were injected intraperitoneally with pregnant mare serum gonadotropin (5 U) and with hCG (5 U) 48 hours later. Error bars represent SEM. (C-E) Distributions of developmental stages of fertilized eggs are shown at each time point (E1.5, E2.5, and E3.5 = days of pregnancy). Embryos were flushed with M2 medium (Sigma) from oviduct or uterus under the natural ovulation condition and counted at the indicated stage. Error bars represent SEM of three to five different experiments; E cell stage: P<.005; E3.5 abnormal: P<.005; E3.5 blastocyst: P<.01.
