Figure 1 IL-8 regulatory region polymorphisms. (A) Sequence analysis of the IL-8 gene 5′-flanking region. Genomic DNA from selected individuals was cloned and sequenced to illustrate the single nucleotide polymorphisms T-845C, T-738A, and A-353T. Representative electropherograms demonstrating the sequence immediately surrounding each polymorphic allele (arrow) are shown. (B) Restriction analysis for IL-8 genotyping. The SNPs at positions -845, -738, and -353 are located in Vsp I, Xba I, and Mfe I sites. These restriction sites are intact if the wild-type alleles are present, but are disrupted if the variant alleles are present. The ethidium bromide-stained gel shows the banding pattern for each SNP after digestion of genomic DNA from individuals homozygous for the wild-type allele or heterozygous for the variant allele.
