The dog was stably engrafted but unfortunately developed severe pancreatitis, a complication occasionally observed in dogs after 920 cGy TBI. The other two dogs (G179 and G197) were transferred to another study evaluating MGMT-mediated in vivo selection and received a drug regimen of O6-benzylguanine and temozolomide (manuscript in preparation). Both of these dogs are currently doing well and continue to show multilineage marking equivalent to or higher than marking levels reported here for the first 100 days after transplantation, with a maximum follow-up of 449 days (G179) and 352 days (G197) after transplantation. (d) Gene marking in subsets of peripheral blood leukocytes and bone marrow CD34+ cells in dogs G179 and G197. Antibodies used for subset staining: anti CD3 (T-cells); anti CD21 (B-cells); anti CD14 (monocytes); DM5 (granulocytes); anti CD34 (bone marrow CD34-positive cells). (e) Gene marking in red blood cells and platelets in dogs G179, G194, and G197. Gene marking was determined by flow cytometry. Gene marking in leukocyte subsets (d) and red blood cells and platelets (e) was determined between day 93 and day 130 after transplantation.
