Hairpin & reporter systems. (A) Positions and sense/antisense orientation of dsRNA and reporter sequences relative to mRNAs. Stem loop structures indicate situations at stem termini. (B) Reporter system performance using various “neutral” co-expressed proteins. 3T3 cells were transfected with MosIR and luciferase reporters and one of EGFP-, RFP-, and LacZ-expressing plasmids. Luciferase activities were analyzed 48 h after transfection. Firefly and Renilla graphs show raw luminescence values from one transfection experiment performed in triplicates, the right graph depicts data from four independent experiments performed in triplicates.
