Figure 1 Macroscopic appearance of laterally spreading tumors (LST) observed with chromoscopic colonoscopy. Representative endoscopic pictures and schematic images of (A) granular-type laterally spreading tumors (G-LST) and (B) flat-type laterally spreading tumors (F-LST) are shown. LST were characterized by a size of more than 10 mm in diameter, a low vertical axis, and lateral extension along the luminal wall. G-LST were composed of superficially spreading aggregates of nodules, forming a flat-based lesion with a granulonodular and uneven surface. F-LST had a flat smooth surface, with an absence of granulonodular formations. Figure 2 Analysis of the methylation status of adenomas by methylation-specific PCR (MSP) and combined bisulfite restriction analysis (COBRA). For analysis of MSP, bisulfite-modified adenoma DNA was amplified in separate reactions using primers specific for unmethylated (U) and methylated template (M). The results of COBRA (C; postdigestion by methylation-specific enzyme) were also presented beneath the corresponding results of MSP. Representative results for p16 and MINT1, 2, 12, and 31 loci are shown. P1 and LG1-3 are determined to be CIMP-high adenomas, with both MSP and COBRA. In most cases, similar results were obtained between MSP and COBRA. However, COBRA was slightly less sensitive than MSP (eg, P1 for MINT31 and LG2 for MINT 12). P1, protruded-type colorectal adenoma tissue samples; LG1-3, G-LST colorectal adenomas; N, normal colon mucosa; R, RKO (colon cancer cell line), W, H2O.
