3 Fig. 2 Schematic representation of IL-4 and IL-13 receptors and signaling pathways. The type II IL-4 receptor is also the functional receptor for IL-13 and is comprised of IL-4Rα and IL-13Rα1. Signaling is mediated predominantly through IL-4Rα and results in activation of the JAK/STAT and IRS-1/IRS-2 pathways. IL-13Rα2 binds IL-13 with high affinity but does not signal and has been hypothesized to act as a decoy receptor. SH2-domain containing tyrosine phosphatases (SHP) downregulate IL-13 signaling by dephosphorylating activated JAKs and STATs. B, SOCS. Expression of SOCS proteins is induced by cytokine, and then SOCSs bind and inhibit JAKs. They might target the JAKs for proteasomal degradation. C, PIAS. Although no PIAS has been yet described for STAT6, PIAS proteins bind to activated STAT dimers and inhibit DNA binding. The schematic overview is based on published observations with STAT1.
