Cellular analysis ofEpotransgenic mice. (A) rHuEPO rescue in transgenic Epo mice. rHuEPO was injected at either 120 U/kg (low dose) and 1200 U/kg (high dose) into EpoWT/WT and EpoΔ/Δ mice. At 4 days after injection, whole blood was measured for reticulocytes by thiazole orange staining. Experiments were performed in triplicate. (B) Hematocrit readings of transgenic mouse after pellet implantation. EpoΔ/Δ and EpoWT/Δ, with or without the EIIa-Cre allele, were analyzed over a 140-day time course. n=6 EpoΔ/Δ; n=5 EpoWT/Δ; n=11 EpoWT/floxCreNeg.
