Quantification method for the nucleotide occlusion assay. The total radio signal of ATP and ADP in the load (before spin-column) corresponds to the total amount of ATP in the sample, whereas the individual signals in the eluate (after spin-column) correspond to the amounts of nucleotides occluded by ABCE1. In addition, the relative amount of ABCE1 in the eluate was determined by SDS–PAGE to exactly adjust the calculation. Representative set of two independent experiments.
