Catalytic site II mutants of ABCE1 show a dominant negative growth effect. In minimal medium lacking histidine (–HIS) and 5-FOA, yeast cells express WT and mutant ABCE1, both under the control of the endogenous promoter. Toxic ABCE1 mutants E493A, E493Q (site II), E247A/E493A (both sites), S223R (site II), and S469R (site I) poise the translation apparatus and cause a dominant negative growth defect even in the presence of the WT gene. In the presence of 5-FOA, the URA3 selection marker leads to loss of the pRS426 vector carrying the WT ABCE1 gene and yeast cells can only survive with the ABCE1 on pRS423. Here, only the cells that rely on the catalytic site I variant E247A are viable. Cells that depend on E247Q show an impaired growth. Elina Nürenberg-Goloub et al.
