Fig. 1 Histological images of the lungs of hamsters instilled and those not instilled with papain to induce emphysema. (A) Control+saline (CS): animals instilled with approximately 0.3mL of saline; (B) emphysema 20mg/mL (E20): animals instilled with approximately 0.3mL of 20mg/mL papain in saline; and (C) emphysema 40mg/mL (E40): animals instilled with approximately 0.3mL of 40mg/mL papain in saline. Animals were euthanized after 60 days. Magnification: 50× (HE). Alveolar destruction was determined by the number of times that a predetermined group of coherent lines (1.25mm2 area and 1.50mm total length) crossed parenchymal structures. The lower the number of times that structures were crossed, the more extensive the lesion. Pathophysiology , DOI: ( /j.pathophys ) Copyright © 2013 Elsevier Ireland Ltd Terms and Conditions

Fig. 2 Right and left ventricle lipid hydroperoxides and antioxidant capacity in emphysema hamsters. (A) The tert-butyl hydroperoxide-induced chemiluminescence was monitored continuously for 40min. The area under the curve for each animal curve was determined to compare groups. (B) Effect of emphysema on total antioxidant capacity in the supernatant of the right and left cardiac muscles of hamsters treated with 2 different doses of papain. Results are expressed in μM trolox. Both analyses were performed by 1-way ANOVA, followed by Bonferroni as the post-test. A value of p<0.05 was considered significant. RV, right ventricle; LV, left ventricle; CS, group instilled with 300μL of NaCl 0.9%; E20, group instilled with 300μL of papain at 20mg/mL in NaCl 0.9%; E40, group instilled with 300μL of papain at 40mg/mL in NaCl 0.9%. Fig. 3 Effect of emphysema on TBARS and carbonyl proteins in the right and left ventricles of hamsters. (A) TBARS levels in heart ventricle homogenates of hamsters instilled with 2 different doses of papain. (B) Levels of protein carbonylation in RV and LV homogenates of control and emphysema hamsters. Results represent means±SD (n=8). For both graphs, RV, right ventricle; LV, Left ventricle. No statistically significant differences were detected by 1-way ANOVA followed by the Bonferroni multiple comparison test as the post hoc test. CS, group instilled with 300μL of NaCl 0.9%; E20, group instilled with 300μL of papain at 20mg/mL in NaCl 0.9%; E40, group instilled with 300μL of papain at 40mg/mL in NaCl 0.9%. Fig. 4 Proteasomal proteolytic activity assessed by chymotrypsin-like activity in the right and left ventricles of hamsters instilled intratracheally and of those not instilled with papain. RV, right ventricle; LV, left ventricle. Statistically significant differences are indicated on the graph as detected by 1-way ANOVA followed by the Bonferroni's multiple test comparison. CS, group instilled with 300μL of NaCl 0.9%; E20, group instilled with 300μL of papain at 20mg/mL in NaCl 0.9%; E40, group instilled with 300μL of papain at 40mg/mL in NaCl 0.9%.
