Loss of Arp2/3 or SCAR activity does not modify α-Cat phenotype during oogenesis. (A–D) Cells mutant for Sop2 and SCAR or single and double knockdown cells compromised for Sop2, SCAR, or α-Cat are marked by the absence of GFP (blue). Cells are labeled for DEcad or α-Spectrin (α-Spec). (A) Loss of Sop2 causes apical constriction and membrane ruffling at AJs in FE cells at stage 11. (B) FE cells expressing Sop2-RNAi constrict apically and show minor membrane ruffling compared to Sop2 mutant cells. (C,D) SCAR mutant cells show variable junctional defects (weak phenotype shown in C and a strong phenotype shown in D) similar to those seen in Sop2 compromised cells. (E) FE cells expressing SCAR-RNAi undergo apical constriction (left side of dashed line is a basal view of the clone and the right side is an apical view). (F) Expression of α-Cat-RNAi causes loss of AJs and formation of α-Spectrin aggregates that are also enriched in DEcad.
