AMPK-mediated mTORC2 signaling does not require mTOR Ser1261phosphorylation. (A) Two littermate-matched pairs of WT (mTOR+/+) and mTOR S1261A (mTORA/A) primary MEFs were serum-starved (20 hours) and treated without (−) or with (+) AICAR (2.5 mM; 2 hours). Whole-cell lysates were immunoblotted as indicated. Blots are representative of four independent experiments. (B) Primary MEFs from mTOR+/+ or mTORA/A littermates were serum-starved (20 hours) and refed with DMEM containing (−) or lacking (+) glucose (8 hours) to induce glucose deprivation (Glc W/D; glucose withdrawal) without or with torin1 (100 nM).
