Lanes 2 through 6 show results obtained by using serial 3-fold dilutions (from 300 pg to 3 pg) of truncated IS cRNA as input. Lanes 7 through 11 are results obtained when a fixed level of test input RNA from a sample of sinus mucosa is incubated with the same range of dilutions of IS cRNA. Lane 12 shows the results when the same amount of test input RNA was subjected to RT-PCR in the absence of any internal standard. Product from authentic IL-8 mRNA is seen at 284 bp, whereas product from the IS cRNA is seen as a band at 189 bp. Competition is observed between the test RNA and the IS. In this instance equivalence was calculated to occur at an IS cRNA level of 22 pg (between the 30 pg and 10 pg levels of IS in lanes 9 and 10 ).
