Fig. 1 Proliferative responses of IELs to various concentrations of IL-2 (▴), IL-7 (■), and IL-15 (●) were measured after 3 days. At concentrations of ≥0.1 ng/mL, IL-15 effected a greater response than IL-2 or IL-7 (n = 5; P < 0.05). Bars represent SEM. Fig. 2 Proliferative responses of CD4+ (□) and CD8+ (-) T-cell subsets were compared with those of unseparated (▨)(A) IELs and (B) LPLs after a 3-day culture with medium, IL-2, IL-7, and IL-15 (10 ng/mL). Fig. 4 IELs, cultured for 3 days with various concentrations of (A) IL-2 or (B) IL-15, were tested for cytotoxic activity against HT-29 colon cancer cells using a range of effector-to-target cell ratios. Concentrations (ng/mL) are as follows: ■, 100; ▴, 10; ▾, 1.0; , 0.1. Representative of 3 experiments. Fig. 6 Various numbers of IELs, cultured with IL-2 (■) or IL-15 (▴) with feeder cells for 10 days, were tested for cytotoxic activity against HT-29 target cells to obtain a limiting dilution analysis of this lytic activity. A plot of the percentage of negative wells at each cell number vs. the number of cells added allowed a linear regression from which the 37% negative cell number could be interpolated. The frequency of cytotoxic precursors was determined by the reciprocal of the number of cells yielding 37% negative wells. Representative of 3 experiments.
