Overview of HEAT-motif-containing proteins. (A) Sequence alignment of HEAT motifs in mouse importin β. A consensus sequence is shown at the bottom. A HEAT motif is composed of a pair of α-helices (A- and B-helices) that is connected by a short linker (turn). Conserved hydrophobic residues in the helices are marked by an orange background. Proline and positively charged (arginine or lysine) residues conserved in the A- and B-helices, respectively, are boxed. (B) In a HEAT motif, the A- and B-helices are arranged in an antiparallel fashion through hydrophobic interactions. Hydrophobic residues are marked in orange. (C) Domain organizations of the three groups of HEAT-motif-containing proteins in humans. HEAT motifs, IDRs (brown) and other functional domains (pink) are shown. HEAT motifs whose crystal structures have been determined are shown in green, whereas HEAT motifs predicted solely based on their primary sequences are shown in gray. Shige H.
