Mouse morulae (A, B) were incubated (24 hours; 37°C) to the blastocyst stage (C–F) without (C, D) or with (E, F) recombinant heparanase (0.1 mg/mL). Embryos were fixed and incubated with polyclonal anti-heparanase antibodies, followed by Cy-2 conjugated goat anti-rabbit antibodies, as described in Materials and Methods. The embryos were then mounted and subjected to confocal microscopy (×100). 3 FIGURE 2 Heparanase activity in blastocysts preincubated with recombinant heparanase. Morulae were incubated (24 hours; 37°C) with recombinant 65-kDa heparanase (•) or medium alone (○). The blastocysts were then washed and homogenized, and the supernatant fractions, equilibrated for protein content, were incubated (16 hours; 37°C, pH 5.8) with sulfate-labeled ECM. Sulfate-labeled material released into the incubation medium was subjected to gel filtration analysis, as described in Materials and Methods. Low molecular weight HS degradation fragments (peak II) were detected only in blastocysts (n = 114) that were first incubated with recombinant heparanase.
