The chemokine receptor 5 Δ32 mutation is associated with increased renal survival in patients with IgA nephropathy  U.L.F. Panzer, Andr Schneider, Oliver M. 2 Figure 1 CCR5 genotype analysis. Example of genotype analysis to determine the presence of the CCR5 Δ32 mutation in patients with IgA nephropathy. The length of the amplified fragment is 215bp for the wild-type and 183bp for the CCR5 Δ32 allele. The polymerase chain reaction (PCR) fragments were separated on a 1.5% agarose gel and patients classified as wild-type (+/+), heterozygous (+/-), and homozygous (-/-) carrier. 3 Figure 2 Influence of the CCR5 Δ32 genotype on renal survival in patients with IgA nephropathy. Kaplan-Meier plots of renal survival are shown in months from the date of biopsy until either end stage renal disease or the last follow up (CCR5 Δ32 genotype vs. wild-type, P < 0.001, Log-rank test). 4 Figure 3 Functional effects of CCR5 Δ32 mutation on chemotaxis. RANTES (100ng/mL), MIP-1α (100ng/mL), MIP-1β (100ng/mL), and MCP-1 (100ng/mL) induced chemotaxis on freshly isolated monocytes was assessed in a chemotactic assay. The number of migrated M/M under basal conditions was standardized as 100%. Chemotaxis of monocytes toward all tested chemokines was significantly increased compared to basal conditions in all three groups with the exception of Δ32/Δ32 vs. basal in response to MIP-1β. Significance of differences between groups is indicated as follows: #P < 0.01+/+ vs. Δ32/Δ32; *P < 0.01+/+ vs. +/Δ32; †P < 0.01+/Δ32 vs. Δ32/Δ32.
