In (A) and (B), a biotin end-labeled DNA oligonucleotide was immobilized onto a streptavidin sensor chip, followed by association of either SSB (left) or SSB-113 (right). Solutions of either χ (A), or primase (B) were passed over the sensor chips at either 24°C (dashed) or 37°C (solid). (C) Primase was passed over immoblized χ at 24°C. (D) χ was passed over immoblized primase at 24°C. Mixtures of 32P-primase and χ were incubated with SSB immobilized to the wells of a 96-well plate and then washed prior to autoradiography. The scheme at the top illustrates the experiment in which 3H-primase is used with Pol III* (plus or minus χ)β, and DnaB in rolling circle assays. Reactions were analyzed for total synthesis (A), and the 3 min reaction was anaylzed in an alkaline agarose gel (B). Primase was added or omitted as indicated. 8 Figure 7 The Primase–Polymerase Switch during Lagging Strand Synthesis
(A) DnaB helicase encircles the lagging strand and primase has synthesized a primer. The holoenzyme consists of a dimer of τ that binds two polymerase cores, one γ complex clamp loader, and two β clamps. τ and primase interact with DnaB. Primase must contact SSB to remain on the RNA primer. (B) The χ subunit of γ complex interacts with SSB, severing the primase–SSB contact and resulting in primase displacement. (C) Primase is then free to synthesize another RNA primer upon contact with DnaB. Diagram (B) also shows that the lagging strand polymerase releases the β clamp and DNA upon finishing an Okazaki fragment. Diagram (C) shows that after γ complex assembles the new β clamp on the upstream primer, the lagging polymerase recruits the new β clamp (shaded dark) assembled on the upstream RNA primer for the next Okazaki fragment.
