5 Figure 4 Acetaldehyde (AcCHO) requires c-Abl to inhibit PPAR-γ function through phosphorylation. Isolated human hepatic stellate cells were transduced with adenovirus (pAd) constructs containing β-galactose (β-gal) as a control, wild-type c-Abl (c-Ablwt), and kinase-dead c-Abl (c-AblKD). Cells were then transfected with a vector containing PPAR-γ (pcmx-PPAR-γ) and incubated with [32P]orthophosphate for 3 hours. Cells were then treated with AcCHO for 15 minutes, and assayed for phosphorylated-PPAR-γ (p-PPAR-γ), which has reduced transcriptional activity. Note that AcCHO cannot induce inhibitory phosphorylation of PPAR-γ with the kinase dead form of c-Abl.
