Figure 1 A typical immunoblot study of rat renal microsomal protein isolated with antibody AS 31.7 and probed with antibodies directed against HKα2 or Na+-K+-ATPase α1(N = 5). The HKα2 protein was isolated from 125 μg of renal microsomal protein from low K (lanes 3 and 4) or normal animals (lanes 5 and 6) using AS Distinct bands were noted with low K and normal kidney probed with AS 31.7 but not with the antibody directed against Na+-K+-ATPase α1. On the other hand, a band was noted when purified renal Na+-K+-ATPase was probed with the Na+-K+-ATPase α1-specific antibody but not with the HKα2-specific antibody. Figure 2 A typical immunoblot study of renal microsomal protein isolated with AS 31.7 and probed with HKβ1-specific antibody or Na+-K+-ATPase β1-specific antibody (N = 6). The HKα2 protein was isolated from 500 μg (lanes 5 and 6), 250 μg (lanes 7 and 8) and 125 μg (lanes 9 and 10) of renal microsomal protein using AS 31.7 and then probed with HKβ1-specific antibody (2B6) or Na+-K+-ATPase β1-specific antibody. Distinct bands were noted with the Na+-K+-ATPase β1 antibody at all concentrations of renal microsomal protein (lanes 6, 8, 10) that comigrated with Na+-K+-ATPase probed with this antibody (lane 4). No bands were detected at any concentration of renal microsomal protein probed with the HKβ1 antibody. Moreover, a band was detected with the G1 fraction of the stomach (purified H+-K+-ATPase) (lane 1) probed with the HKβ1 antibody but not when the G1 fraction was probed with Na+-K+-ATPase β1 antibody.
