1 FRDC : Aquatic Animal Health Subprogram: Determining the susceptibility of Australian Penaeus monodon and P. merguiensis to newly identified enzootic (YHV7) and exotic (YHV8 and YHV10) Yellow head virus (YHV) genotypes. AUSTRALIAN ANIMAL HEALTH LABORATORY (AAHL) Nick Moody | Team Leader, Aquatic Diagnostic Capability Ridley Australian Prawn & Barramundi Farmers Symposium – 2 August 2016

2 Project Update YHV infectivity trials

3 FRDC 2015-015 PI: Nick Moody, AFDL, Geelong
CI: Mark Crane, AFDL, Geelong Start Date: End Date: FRDC funds: $159,941 Objectives No. Details 1 Determine the susceptibility of P. monodon and P. merguiensis to YHV-complex Genotype 7 (YHV7) 2 Determine the susceptibility of P. monodon and P. merguiensis to YHV-complex Genotype 8 (YHV8) 3 Determine the susceptibility of P. monodon and P. merguiensis to YHV-complex Genotype 10 (YHV10) 4 Transfer protocols and controls for diagnostic tests to state diagnostic laboratories YHV infectivity trials

4 FRDC 2015-015 Infectivity trial: November, 2015
1: Sequenced as Wenzhou shrimp virus-1 2: Sequence similarity with HPV2 and HPV3 from Madagascar 3: Sequence similarity of 96% with Australian HPV YHV infectivity trials

5 FRDC YHV1 is pathogenic to P. monodon and P. merguiensis by inoculation YHV infectivity trials

6 FRDC 2015-015 YHV7 is pathogenic to P. monodon by inoculation
YHV infectivity trials

7 FRDC YHV8 is not pathogenic to P. monodon and P. merguiensis by inoculation YHV infectivity trials

8 FRDC YHV10 is not pathogenic to P. monodon and P. merguiensis by inoculation (inoculum also had YHV8 in it) YHV infectivity trials

9 FRDC YHV infectivity trials

10 Conclusions Demonstrated pathogenicity of YHV1 to Australian P. monodon and P. merguiensis (by inoculation) Demonstrated pathogenicity of YHV7 to Australian P. monodon (by inoculation) YHV2 (GAV) in host P. monodon and YHV7 inoculum did not appear to have any effect on the prawns (temperature too low: 24  1C?) Generated material for feeding and co-habitation trials Redo YHV10 inoculation with additional material from imported prawns obtained after the November 2015 trial Seeking prawns for infectivity trials in October/November 2016 YHV infectivity trials
