In situ footprinting of the IGRP promoter: primer set D. Y1 (lanes 1 and 2) and βTC-3 (lanes 3 and 4) genomic DNA was methylated in vitro (lanes 1 and 3) or within intact cells in situ (lanes 2 and 4), using DMS (as described in research design and methods). After cleavage of methylated DNA with piperidine, an LMPCR reaction was performed using IGRP primer set D (Table 1). The autoradiograph shown is representative of two experiments. Only increases (•) or decreases (○) in band intensity, comparing in situ to in vitro methylated DNA, that were consistent between both gels are indicated. Larry J. Bischof et al.
