Fig. 1 Time course of expression of gelatinases after injury. Gelatin zymogram shows 72 kd gelatinase was constitutively expressed in uninjured carotid artery (C). Between 5 and 14 days (d) after injury, activities at 59 kd and 55 kd were increased. A 92 kd gelatinase was absent in controls, was significantly increased 1 day after injury, and gradually decreased thereafter. There was low molecular weight gelatinolytic activity at 24.5 kd that increased 5 days after injury. Fig. 2 Gelatinolytic activity in thickened intima versus adventitia at 14 days after injury. Activity of 72 kd gelatinase in intima was greater than in adventitia. Activity of 92 kd gelatinase was faint and about same in both tissues, whereas gelatinolytic activity at 24.5 kd was present only in adventitia. Fig. 3 Time course of expression of caseinolytic proteinases by α-casein zymography. Caseinolytic activities at 27 kd and 24.5 kd were detectable in uninjured carotid artery (C). At 5 days (d) after injury, activity at 24.5 kd was significantly increased and remained elevated for 14 days. Low molecular weight caseinolytic activity was much less in intima than in adventitia Journal of Vascular Surgery , DOI: ( / (94) ) Copyright © 1994 Society for Vascular Surgery and International Society for Cardiovascular Surgery, North American Chapter Terms and Conditions

Fig. 5 Soluble elastin zymogram demonstrates elastolytic activity at 24.5 kd in carotid artery extract (1) and adventitial extract (2). One hundred micrograms of protein were loaded per lane. Fig. 6 Expression of 24.5 kd caseinase in adventitial and lumenal extracts compared with cultured cells. Caseinolytic activity at 24.5 kd was much higher in adventitial extract (1) than in aortic lumenal extract (2) (10 μg of protein loaded per lane). Conditioned medium (40 μl per lane) from cultured SMCs (3), adventitial fibroblasts (4), and capillary endothelial cells (5) had no caseinolytic activity at 24.5 kd. However, conditioned medium from peritoneal macrophages (6) did show caseinolytic activity at 24.5 kd.
