Biphasic increase of PI(4,5)P2 level in LPS-stimulated cells. J774 cells were stimulated for the indicated times with 10 ng/ml (A) or 100 ng/ml (B,C) of LPS with or without pre-incubation with 10 μg/ml of a function-blocking antibody against CD14. After extraction of phosphoinositides, PI(4,5)P2 amounts in samples were estimated using ELISA (A,B) or a protein–lipid overlay assay using the PLC-PH–GST probe, followed by densitometry analysis (presented in the histogram, C). (C, right) The results of the protein–lipid overlay assay, which was performed on lipids that had been extracted from unstimulated cells or cells stimulated with 100 ng/ml LPS for 5 and 60 min. Samples were incubated with the PLC-PH–GST probe, which had been pre-adsorbed with liposomes containing or devoid of PI(4,5)P2.
