In the absence of ligand, Sam68 interacts with FADD. Upon activation with TNF-α, Sam68 is recruited to TNFR1, where it stabilizes cIAP1 and TRAF2, allowing RIPK1 ubiquitylation and NF-κB activation. TNFR1 is internalized; RIPK1 and Sam68 disassociate from the receptor and form a complex with FADD and caspase-8, leading to caspase activation and cell death.
