CD27+ expression correlates with robust Ag-specific responses to M. bovis. Approximately 8 wk after virulent M. bovis infection, PBMCs (5 × 106 cells/ml) were labeled with CellTrace Violet and restimulated in vitro for 6 d in the presence or absence of PPD-b, ESAT-6/CFP-10 or left unstimulated. Cells were then labeled with anti-bovine CD4 or γδ TCR, and CD27. CellTrace dilution (proliferative responses) and CD27 cell surface expression was determined for (A) γδ T cells and (B) CD4 T cells (Supplemental Fig. 2).
