The expression of core clock genes under normal or UVR-irradiated conditions. Human epidermal keratinocytes were synchronized with serum-rich medium for 2 h and exposed to UVB (20 mJ/cm2) at ZT 2 (A), ZT 8/ZT 32 (B, C) or ZT 20/ZT 44 (D). The mRNA expression of CLOCK (A, B) and BMAL1 (C) was analyzed by qRT-PCR. The data are presented as the mean ± SD of two independent experiments. Con, non-treated cells; UVB, UVB-treated cells. (D) The protein expression of CLOCK and BMAL1 was examined by western blot analysis. GAPDH was used as a loading control. Densitometry analyses of BMAL1 protein expression over GAPDH control were performed using ImageJ program and the quantified data are presneted as the mean ± SD of thtree independent experiments (histogram).
