Figure 1 Cholesterols and Membrane Composition Drive ISC Proliferation and Tumorigenesis (A) In Lpcat3 wild-type mice (WT), active phospholipid-remodeling enzyme Lpcat3 maintains membrane integrity, basal cholesterol biosynthesis, and proliferation in intestinal crypt cells and tumors. Dietary cholesterols promote the proliferation of both intestinal crypt cells and tumors. (B) In Lpcat3 knockout mice (KO), Lpcat3 deficiency impairs phospholipid remodeling and induces cholesterol biosynthesis, leading to hyperproliferation of intestinal crypt cells and tumors. Ro (Ro48, LSS inhibitor) blocks cholesterol biosynthesis and reduces the hyperproliferation of intestinal crypt cells and tumor growth.
