(A) Lalit et al. show that overexpression of five cardiac genes in mouse fibroblasts with BIO and LIF promote emergence of iCPCs that express a distinct set of CPC markers. These cells can be expanded >1015-fold and differentiate into cardiomyocytes (CMs), endothelial cells (ECs), and smooth muscle cells (SMCs) and contribute to the heart and promote survival after their transplantation into embryos and after myocardial infarction (MI). (B) Zhang et al. show that Yamanaka factor expression with JAK inhibitor JI1 and BACS (BMP4, Activin, CHIR99021, and SU5402) reprograms mouse fibroblasts into ieCPCs with large expansion capacity. ieCPCs differentiate into CMs, ECs, and SMCs in vitro and after transplantation into MI hearts, where they provide functional benefit.
