1 The CACTA Transposon Is Hypermethylated in U-S Plants.
The CACTA Transposon Is Hypermethylated in U-S Plants. (A) Bisulfite sequencing (BS-seq) of the left border region of the CACTA and the flanking sequence is shown by a line below this region. The gray box represents the CACTA transposon, and black boxes indicate the transcribed regions (5′ UTR) of the Ufo1-1 transcript. Coordinates shown are relative to the start of the CACTA. U-E-revertant plants are the progeny of U-S plants that have regained some flavonoid pigment expression. Data are averages of two independent plants per genotype, with error bars representing the se. A minimum of 22 clones were sequenced per replicate. TSS, transcription start site. (B) sRNA (nucleotides 20–24) abundance throughout the CACTA shown as the average of three biological replicates per genotype. (C) qRT-PCR of Ufo1-1 candidate GRMZM2G and reporter p1 from mature leaves, 10-DAP developing seed, 18-DAP pericarp (pc), 18-DAP endosperm (endo), and 18-DAP embryo (emb). Expression was calculated relative to actin1 as the average of three biological replicates, with error bars representing the se. Kameron Wittmeyer et al.
