(A), Northern blot analysis of LH/hCG receptor, AM, and RAMP2 mRNA in human corpus luteum during the menstrual cycle and early pregnancy. Ten micrograms of total RNA from individual samples were fractionated on an agarose gel, transferred to membranes, and hybridized by using the cRNA probes. These data represent the results of four experiments: follicle (F) (cycle day 14, 20 mm in diameter), mid-luteal phase (L) (cycle day 24), pregnancy at 7 weeks P7w, and pregnancy at 13 weeks (P13w). (B), Autoradiographs of RAMP2, AM, and LH/hCG receptor (5.4 kilobases) mRNA were quantified by using densitometric scanning. The amount of RAMP2 mRNA in the follicle was assumed to be 100% and the amounts of AM and LH/hCG receptor mRNA in the mid-luteal phase were assumed to be 100%. Data were normalized for levels of glyceraldehyde-3-phosphate dehydrogenase mRNA levels in each sample and are expressed relative to the control value (mean±SE; n=3). ∗ Difference from the control value at P<.05.
