Defective immune responses in mice lacking LUBAC‐mediated linear ubiquitination in B cells
Cpdm mice show residual LUBAC‐mediated linear polyubiquitination activity. (A) Lysates of splenic B cells from control, B‐HOIPΔlinear, or cpdm mice were immunoblotted with the indicated antibodies. Data are representative of three independent experiments. (B) Splenic B cells were stimulated with 5 ng/ml of CD40L for the indicated times and then immunoblotted with the indicated antibodies. Data are representative of three independent experiments. (C) Splenic B cells were stimulated with an anti‐CD40 antibody (3 μg/ml) for the indicated times. Whole‐cell lysates (WCL) were prepared and immunoprecipitated with an anti‐NEMO antibody and then immunoblotted with an anti‐linear ubiquitin antibody (LUB9) (upper panel). WCL were also immunoblotted with an anti‐NEMO antibody (lower panel) to confirm that the lysates contained equal amounts of NEMO protein.
