FIGURE 1 Spontaneous secretion of MIF (A) and MCP-1 (B) by peritoneal macrophages from control women (N) and those with endometriosis (E). Macrophages were isolated from the peritoneal fluid, resuspended in RPMI medium and dispensed in 24-well culture plates at 1 × 106 cells per well. Adherent cells were rinsed and incubated with 1 mL per well of RPMI for different time periods. Macrophage migration inhibitory factor and MCP-1 concentrations in the culture supernatants were measured by ELISA. Data were expressed as mean ± SEM of triplicate values in 12 healthy women and 11 women with endometriosis. ∗Statistically significant difference between healthy subjects and those with endometriosis (P<.05). Peritoneal macrophages, prepared as indicated in Figure 1’s legend, were incubated with 1 μg/mL of LPS (+LPS) or with the culture medium alone (−LPS). Six hours later, the culture supernatants were recovered, and the concentrations of MIF and MCP-1 were measured by ELISA.
