Goldrath and colleagues utilize VHL-deficient T cells, which express persistently stable HIF1α, to enforce constitutive glycolysis in CD8+ T cells and inhibit mitochondrial activity and development of spare respiratory capacity (SRC). These T cells maintain the ability to form memory in response to acute viral infection but show increased proportions of “effector memory” T cells. This demonstrates that T cells can utilize many metabolic pathways to differentiate into the memory phase and that metabolic pathways may be more important in licensing rather than dictating T cell fate decisions.
