FIGURE 1 Representative Southern blot assay of aromatase cytochrome P450 (P450arom) transcripts after amplification with reverse transcription polymerase chain. In the in vivo study, endometrial specimens were obtained from patients with endometriosis, adenomyosis, or leiomyomas before and after 6 months of treatment with gonadotropin-releasing hormone agonists (GnRHa) or danazol. In the in vitro study, the endometrial explants obtained from untreated patients with endometriosis, adenomyosis, or leiomyomas were cultured for 48 hours in the absence (control) or presence of GnRH agonist or danazol. Total RNA were reverse transcribed, amplified with aromatase cytochrome P450 or glyceraldehyde-3-phosphate dehydrogenase (G3PDH) primers, and hybridized with the 32P-labeled corresponding probes, as described in Materials and Methods. bp = base pairs.
