S-Affs colocalize with SUMO in mammalian cells.
S-Affs colocalize with SUMO in mammalian cells. (A) Immunofluorescence showing FLAG-S-Affs expressed in HEK293T cells. S-Affs that colocalized with structures resembling nucleoli are denoted with white arrowheads, and those localizing with structures resembling PML-NBs are denoted with white arrows. 4′,6-Diamidino-2-phenylindole (DAPI) stain (blue) labels the nuclei. Images shown are representative of three independent experiments. Scale bar, 20 μm. (B) Quantification of FLAG-S-Affs colocalization with nucleoli in untreated cells and in cells treated with arsenic trioxide [As2O3 (As)]. The average percent of S-Aff–positive cells displaying nucleolar staining was derived from five independent images per experiment (n = 3), with about 70 to 150 positive cells per image. Error bars represent the SD from three independent experiments. (C and D) Immunofluorescence showing the indicated FLAG-tagged S-Affs (green) and SUMO-1 (red) (C) or SUMO-2/3 (red) (D) in HEK293T cells treated with As. Control cells were transfected with empty vector. Scale bars, 5 μm. David J. Hughes et al., Sci.
