2 Figure 1 The impact of c-Jun NH2-terminal kinase inhibitor (JNK-I), with or without medroxyprogesterone acetate (MPA), and GnRH antagonist (cetrorelix) treatment on (A) number, (B) volume, and (C) surface area of induced endometriosis lesions. Results are expressed as a ratio: (pre-treatment–post-treatment for drug-treated animals)/(pre-treatment–post-treatment for vehicle treated animals). This ratio is 1 for vehicle-treated animals and was significantly lower in all treated groups for volume and surface area of all, typical, and red lesions and number of red lesions, whereas the ratio for the number of typical lesions was reduced only in the JNKI + MPA group and cetrorelix groups. The paired (*) statistical comparisons were determined from the original measures of number of lesions, surface area of lesions (mm2), and volume of lesions (mm3), with original values (mean ± SD, median, range), presented in Supplemental Table 1 (available online at rASRM score = revised American Society for Reproductive Medicine score. * P< .05 when compared to placebo. 3 Supplemental Figure 1 Exposure profile of AS in baboon plasma after oral administration of either 10 mg/kg or 20 mg/kg in a single administration. Mean (±SD) AS plasma concentration over the first 24 hours is presented in a log-linear scale. Exposure profile at 20 mg/kg per day contained less variability among the baboons and maintained plasma levels of AS above the 50% inhibitory concentration for ≥12 hours.
