Embryos were dissected and opened along the ventral midline (floor plate) before culture. C, D, Illustration of the bead implantation experiments. PBS-soaked (black asterisk) or Fgf8-soaked (red asterisk) beads were inserted into the alar or roof plates of the explanted diencephalon. Twenty-four hours after Fgf8 bead insertion, Wnt3a (C) and Mkp3 (D) expression was induced around the Fgf8 beads in the diencephalon of both normal and Fgf8null/neo embryos. The phenotype of explants was indicated in the corresponding pictures.
