CPEB1 protein interacts with the MTDH/AEG-1 mRNA. A, MTDH/AEG-1 mRNA contains a CPE sequence within 100 nucleotides (nt) of the hexanucleotide (HEX) sequence that is conserved where sequence was available, including humans, non-human primates, bovines, and rodents. Shown are the sequences in humans and mice (accession# NM_178812, NM_026002). B, CPEB1 protein was immunoprecipitated (IP) from CNS1 cells in culture using an antibody specific for CPEB1 but not when a nonspecific immunoglobulin G (IgG) antibody was used. RNA was isolated from the immunoprecipitation and RT-PCR was conducted using primers specific for MTDH/AEG-1 (MTDH) or GFAP (n = 3). C, CNS1 cells were transfected with DN-CPEB1-GFP followed by immunoprecipitated using an antibody specific for GFP. RNA was then isolated and an RT-PCR was again conducted using primers specific for MTDH/AEG-1 or GFAP (n = 3). WB, Western blot analysis.
