The percent activity of each well was calculated relative to podophyllotoxin (defined as 100% activity) and DMSO (defined as 0% activity). The results are depicted here as a gradient of color where white, blue, and red indicate no, increasing, and decreasing activity, respectively, relative to DMSO-treated control wells. These data were converted into dose–response curves. Podophyllotoxin dose–response curves from the qHTS runs (B and C) in which each of 7 plates containing the concentration indicated in (A) are shown for SW480 (B) and MCF10A (C) cells. Intraplate control titrations of 16 different concentrations of podophyllotoxin (D and E) are shown for SW480 (D) and MCF10A (E) cells.
