Hybridomas expressing Vγ1.1Vδ6.3 TCRs also recognize macrophages isolated from the peritoneal cavity, and the response increases when activated with macrophages infected with Listeria. (A) Hybridomas transfected with variants 2-10, 3-10, 6-10, and of the Vγ1.1Vδ6.3 TCR were incubated alone (No Mϕ), in the presence of uninfected (UI) macrophages, or in the presence of macrophages infected with Listeria at an MOI of 0.1, 0.5, 1, and 5. After 16 h of incubation, IL-2 levels in the supernatant were assayed by ELISA. The bars represent the mean of three independent experiments, and the error bar represents the SD of the mean. Statistical analysis was performed using a paired t test. The asterisk above the bracket indicates statistical difference between the sample with No Mϕ and the UI treatment.
