NF-kB binds to the NF-κB binding site immediately adjacent to the AT-rich regulatory region. Electrophoretic mobility shift assay was done by incubating 5 μg nuclear extract of nontransfected (lane 1) or p50/p65–transfected (NIH/3T3+, lanes 2-6) NIH/3T3 cells with biotin-labeled wild-type (lanes 1-5) or mutant (lane 6) Imp2 promoter oligonucleotides. Where indicated, the samples were preincubated with antibody directed against p50 (lane 3) or p65 (lane 4) or with HMGA2-specific antiserum (lane 5).
