Lamellarin D induces cell death through a Fas-independent pathway. A, expression of Fas and Fas ligand (Fas L) in response to lamellarin D exposure. P388 cells were incubated for the indicated times with or without 5 μmol/L lamellarin D, and whole-cell lysates were subjected to immunoblotting for detection of Fas and Fas L. G3PDH served as a loading control. B, wild-type, FADD-deficient and caspase-8 deficient Jurkat T cells were treated with anti-Fas antibody (500 ng/mL) or lamellarin D (5 μmol/L) for 24 h. The cells were stained for sub-G1 and analyzed by flow cytometry. C, lamellarin D induced the mitochondrial pathway of apoptosis in wild-type, FADD-deficient and caspase-8–deficient Jurkat T cells. After 12 h of treatment with 5 μmol/L lamellarin D, cytosolic fractions were analyzed by immunoblotting using cytochrome c antibody and the percentage of cells with a decrease in Δψm (Δψmlow) were measured by flow cytometry.
