Effects of visfatin on the cell proliferation and phosphorylation of ERK, Akt, and GSK-3β proteins in HCC cells. Effects of visfatin on the cell proliferation and phosphorylation of ERK, Akt, and GSK-3β proteins in HCC cells. HepG2 (A), Hep3B (B), and HuH7 (C) cells were treated with the indicated concentration of visfatin for 48 hours in serum-free medium. Cell proliferation was evaluated by an XTT assay. Results were expressed as a percentage of the control value. Bars, SD of triplicate assays. *, P < D, HepG2 cells were treated with and without 100 ng/mL visfatin for 30 minutes, and cell lysates were prepared. The cell lysates were then analyzed with a Western blot using respective antibodies. Equal protein loading was verified by the detection of GAPDH. Repeated Western blotting yielded similar results. p-ERK, phosphorylated ERK; p-Akt, phosphorylated Akt; p-GSK-3β, phosphorylated GSK-3β. Soranobu Ninomiya et al.
