CD4+ T cells and CD8+ T cells isolated from peripheral blood samples were stimulated for 4 h with PMA and ionomycin, and their production of IL-4, IL-13, and IFN-γ was determined at the single cell level by intracellular cytokine staining and flow cytometry as described inMaterials and Methods. Freshly isolated CD8+ CD45RO+ T cells were purified from AD patients with high levels of serum IgE (AD-H) and AD patients with low levels of serum IgE (AD-L). The spontaneous expression of IL-4 and IFN-γ mRNA in cells were determined with RT-PCR as described.
