sir2α null cells differentiate in vitro. RNA was isolated from the differentiating ES cells and separated by electrophoresis on agarose gels before being transferred to membranes for hybridization to various radioactive probes. The probes used reacted with transcripts encoded by the genes indicated on the right. Oct4 is a marker for ES cells while Brachyury and endoA are markers for differentiating cells. The other transcripts (SNRPN, gtl2, lo1) were identified as amongst those absent from the double knockout cells in microarray experiments. GAPDH is the loading control. Michael W. McBurney et al.
