2 Figure 1 Binding of β-Arrestin to the GPCR Results in Receptor Internalization to Endosomes. The β-arrestin-bound GPCR is able to couple to heterotrimeric G proteins within the endosome, forming a ‘megaplex’. Activation of the G protein within the megaplex results in dissociation of the Gβs subunit which can activate endosomal adenylate cyclase leading to the sustained production of cAMP.
